摘要
目的研究金属硫蛋白(metallothionein,MT)、p53蛋白在非小细胞肺癌(non-small-cell-lungcarcinoma,NSCLC)中的表达及其与临床病理的关系。方法选取NSCLC组织73例(鳞癌41例,腺癌32例)及正常肺支气管黏膜组织15例的石蜡标本,用免疫组化技术(S-P法)检测MT及p53蛋白在各组织中的表达。结果MT蛋白和p53蛋白在NSCLC中的阳性表达率分别为54.8%(40/73)、65.8%(48/73),均明显高于在正常肺支气管黏膜组织中的阳性表达率20.0%(3/15)、33.3%(5/15),差异具有统计学意义P<0.05。肺鳞癌和腺癌中MT的阳性表达率分别为56.1%(23/41)和53.1%(17/32),p53蛋白的阳性表达率分别为68.3%(28/41)和62.5%(20/32),其差异均无统计学意义P>0.05。MT蛋白和p53蛋白的阳性表达率均与NSCLC组织的分化程度、淋巴结转移等因素密切相关,差异具有统计学意义(P<0.05)。MT蛋白阳性表达与p53蛋白阳性表达呈正相关(P<0.05)。结论MT蛋白的高表达可能是非小细胞肺癌的发生的一种生物学标志,且与p53蛋白存在协同作用,联合检测两种蛋白的表达水平对判断NSCLC恶性程度、淋巴转移趋势具有重要价值。
Objective To Investigate the expression of metallothionein (MT) and p53 in non-small cell lung carcinoma(NSCLC). Methods The expression of MT and p53 was detected by immunohistochemical method in the paraffin-embedded specimens of 73 NSCLCs and 15 normal bronchial mucosal tissues(NBMT). Results [WTBZ]The positive rates of MT and p53 expression in NSCLC were 54.8%(40/73)and 65.8%(48/73), respectively, and in NBMT were 20.0%(3/15)and 33.3%(5/15), respectively. The positive rates of both MT and p53 in NSCLC were significantly higher than those in NBMT(P<0.05). The positive rates of MT and p53 in lung squamous cell carcinoma were 56.1%(23/41)and 68.3%(28/41), respectively, and in lung adenocarcinoma were 53.1%(17/32) and 62.5%(20/32), respectively. There was not a significant difference in the positive rates of MT and p53 between lung squamous cell carcinoma and adenocarcinoma(P>0.05). The positive rates of MT and p53 were closely correlated with the histological grades and lymph node metastasis of NSCLC(P<0.05). The positive rate of MT was positively related with that of p53(P<0.05). Conclusion MT over-expression may be a biological marker in NSCLC, and was associated with the over-expression of p53. The combination detection of MT and p53 was helpful for evaluating malignant degree and lymph node metastasis of NSCLC.
