摘要 目的建立大鼠新鲜骨组织基因组DNA的提取方法。方法取大鼠肋骨,研磨成粉末,脱钙,水洗,蛋白酶K消化,有机溶剂抽提,离心沉淀干燥,-20℃冰箱中保存。结果提取的骨组织基因组DNA纯度较高,数量大,性质稳定。结论本提取方法,简便易行,获得的基因组DNA量大,纯度高,值得推广。 Objective To establish a method of extracting genome DNA from fresh rat bone. Methods Ratribs were ground to fine powder, decalcified, washed by water and digested by protease K followed by extractionwith organic solvent, centrifugal and sedimentary dehydration and storage at a - 20 ℃ refrigerator. ResultsGenome DNA extracted by the method was of high purity and large quantity, the character of genome DNA wasstable. Conclusions The method is easy to handle, by which genome DNA is extracted in a good amount and ahigh purity. The method is worth of recommendation.
