摘要
目的:研究低密度脂蛋白对体外培养的胰岛细胞功能的影响.方法:体外分离培养Wistar大鼠胰岛细胞,并在低糖(2.8mmol/L)和高糖(16.7mmol/L)条件下与低密度脂蛋白(LDL)(从8mg/L开始)培养48h,应用放射免疫法测定葡萄糖刺激的大鼠胰岛细胞胰岛素分泌量.结果:①低糖刺激下,LDL对胰岛细胞的胰岛素分泌无明显影响,但在高糖刺激下,胰岛素分泌水平降低.②LDL和抗氧化剂维生素E,C共同培养48h,与LDL组比较,高糖刺激下胰岛素分泌水平增加.结论:胰岛细胞摄入LDL后发生氧化反应,损伤胰岛细胞功能,这一过程可被抗氧化剂抑制.
AIM: To study the effect of low density lipoprptein (LDL) on the function of islet cell in vitro. METHODS: Wistar rat islet cells were isolated for cell culture and treated with LDL (from 8 mg/L on) for 48 h. The cells were cultured with low glucose (2.8 mmol/L) and high glucose (16.7 mmol/L). Insulin secretion of rat islet cells stimulated by high glucose was measured by radioemmunoassay. (RESULTS:) ① No obvious effect was observed of the islet cells with LDL at low glucose level, while the insulin levels decreased in rat islet cells cultured with high glucose. ② Compared with LDL, the insulin levels of glucose stimulating insulin secretion (GSIS) increased in rat islet cells cultured with LDL combined with vitamins E and C for 48 h. CONCLUSION: Uptake of LDL by islet cells and subsequent oxidative reactions can damage the cells. This process can be counteracted by antioxidants.
