摘要
目的通过比较幽门螺杆菌(Helicobacterpylori,Hp)感染蒙古沙鼠前后胃粘膜INF-γ、IL-4及IL-12p40mRNA水平的变化,确定Hp感染在蒙古沙鼠胃粘膜诱发的免疫应答方式。方法采用临床分离的Hp强毒株对蒙古沙鼠进行感染,4周后剖杀。取胃粘膜组织作半定量RT-PCR检测INF-γ、IL-4及IL-12p40mRNA。结果与对照组相比,沙鼠感染Hp后,胃粘膜INF-γmRNA水平显著增加(P<0·05),IL-4mRNA水平显著降低(P<0·05),IL-12p40mRNA水平变化不显著(P>0·05)。结论Hp感染沙鼠诱导Th1型免疫应答,同时抑制Th2免疫应答,而对单核巨噬细胞的活化作用不明显。
Objective To analyze the mRNA expressions of INF-γ, IL-4 and IL-12p40 in the gastric mucosa of Mongolian gerbil infected of H. pylori and to investigate the immune response mechanisms. Methods Twenty female Mongolian gerbils of 8 weeks, weighing (50±5) g, were orally administered with H. pylor. Another 20 gerbils were used as control. Four weeks later, all animals were sacrificed and the gastric mucosa was dissected and the mRNA expressions of INF-γ, IL-4 and IL-12p40 were detected by semi-quantitive RT-PCR. Results The mRNA expression of INF-γ increased (P<0.05) but IL-4 decreased significantly (P<0.05), and IL-12p40 was of no change, as compared with the control group. Conclusion H. pylori infection can induce Th1 and depress Th2 response in the gastric mucosa of Mongolian gerbil and has little effect on mononuclear macrophage activation.
