摘要 目的观察鸡胚脊髓开放性神经管缺损(ONTDs)动物模型的组织形态学变化,探讨制作ONTDs动物模型的方法。方法采用显微手术切开神经管,制作鸡胚ONTDs动物模型,再继续孵化3d、5d、7d、10d,观察其组织形态学变化。结果模型制作后不同时间均可见到神经管完全裂开、神经组织外翻、暴露于羊水等典型的ONTDs改变。结论显微手术切开神经管建立鸡胚ONTDs动物模型,方法简单,其组织形态学变化与临床实际神经管缺损(NTDs)相符。 Objective To observe the histomorphological changes of open neural tube defects (ONTDs) in spinal cord of chicken embryo model induced by surgery, and study the method to make the ONTDs animal model. Methods The midline of neural tube between two planes in chicken embryo was incised to induce ONTDs, and the window on the egg shell was sealed by clear tape. All the eggs were then placed in a temperature-controlled incubator until 3, 5, 7, 10 days for the later evaluation of ONTDs development under microscope. Results After ONTDs being surgically induced, complete separation, external rotation of neural tissues exposing to amniotic fluid were seen on all chicken neural tubes in every period, which showed a typical histological character of spinal cord ONTDs; and it is consistent with that in human ONTDs. Conclusion It is simple and efficient to establish ONTDs model by using microsurgical technique to induce ONTDs in chicken embryo, and its morphological character is in conformity to clinical ONTDs.
