甲基强的松龙对预防脊髓损伤的实验研究 被引量:4

摘要 目的探讨预防应用甲基强的松龙(MP)对急性脊髓损伤的影响,为临床上应用药物预防脊髓损伤提供理论指导。方法采用改良Allen’s打击法建立大鼠急性脊髓损伤模型。随机将114只大鼠分为正常组、对照组及甲基强的松龙组,术后采用胥少汀6级行为学评分及Rivlin斜板法对各组大鼠进行脊髓功能评价;对大鼠脊髓损伤前后运动诱发电位的潜伏期以及振幅进行分析;通过光镜和电镜进行病理学观察进行分析。结果1.MP组行为学评分障碍率及斜板障碍率低于对照组(P<0.05),但高于正常组(P<0.05);2.MP组术后MEP潜伏期较对照组短,波幅下降较对照组小(P<0.05),但与对照组无明显差别(P>0.05);3.光镜、电镜下见MP组脊髓损伤轻,神经元细胞正常。结论初步研究表明术前应用MP对脊髓损伤有预防作用。 Objective To study the possibility of the prevention of spinal cor d injury in rat by using Methylprednisolone(MP) before operation of spine,to fin d new treatment in clinic to improve the functional recovery of nerve,also to pr ovide the theoretical guide to apply the drug prevention for spinal cord injury. Methods Build acute spinal cord injury model in rats according to rate by modi fied Allen's method. 114 healthy adult Sprague-Dawley rats were randomly divided into three groups:normal group,control group and Methylprednisolone group. The function of spinal cord was evaluated by Xu shaoding's 6 graded rate and Rivlin' s rate of slope. The changes of the motor evoked potential(MEP)were studied. Nor mal HE and electronmicroscope would be used to observe the pathological and ultr astructural changes of spinal cord. Results The disabled rate of hindlimb locom otor function of Methylprednisolone group is far below that of control group (P< 0.05), but higher than that for normal group(P<0.05). The latent period and ampl itude of MEP for Methylprednisolone group is significantly lower than that for c ontrol group (P<0.05),but the difference is not significant between those two gr oups(P>0.05). Observed under microscope,the spinal cord injury for Methylprednis olone group is not serious. Conclusion Methylprednisolone in prophylaxis can im prove the neurological function of spinal cord.
