摘要
目的:观察当归芍药散(Danggui Shaoyao San,DSS)对β-淀粉样蛋白(Aβ25-35)诱导的PC12细胞损伤的保护作用。方法:制备含DSS脑脊液(CSF-DSS)与含DSS人工脑脊液(ACSF-DSS),应用Aβ25-35诱导PC12细胞损伤,MTT法测定细胞活力和比色法测定细胞内过氧化氢酶(CAT)活性。结果:10μmol/L Aβ25-35可显著降低PC12细胞活力及CAT活性;5 mg/L ACSF-DSS和0.93 g/kg-0.5%、1.86 g/kg-1.0%CSF-DSS均可显著提高细胞活力与CAT活性。结论:DSS可能通过提高CAT活性而对Aβ25-35诱导的PC12细胞损伤起到保护作用。
AIM:To observe the protective effect of cerebrospinal fluid(CSF)and artificial cerebrospinal fluid (ACSF)containing Danggui Shaoyao San(DSS)(CSF/ACSF-DSS) on beta-amyloid peptide fragment 25-35 (Aβ25-35)-induced PC12 cells cytotoxicity.METHODS:CSF-DSS and ACSF-DSS were prepared in rabbits administered with aqueous extract of DSS(ig 0.93,1.86 g/kg),MTT and catalase (CAT) assay were used to measure the viability of PC12 cells.RESULTS:Aβ25-35 (10 μmol/L) caused significant decrease in the viability of PC12 cells 24 h after the treatment.Both ACSF-DSS(5 mg/mL) and CSF-DSS (0.93 g/kg-0.5% or 1.86 g/kg-1.0%) substantially increase the viability and CAT activity of Aβ25-35 (10 μmol/L)-injured PC12 cells.CONCLUSION:DSS could prevent PC12 cells from Aβ25-35-induced cytotoxicity possibly by the mechanism of increasing CAT activity.
