白血病抑制因子在自然流产绒毛组织中的表达 被引量:8

摘要 目的探讨白血病抑制因子(LIF)在绒毛组织中的表达及其与先兆流产及难免流产的关系。方法选择2003年10月~2004年8月来我院就诊、自愿采用人工流产术终止妊娠者为检测对象,按诊断分为正常早孕组、先兆流产组和难免流产组,每组各30例,采肘静脉血并收集流产绒毛组织。采用放射免疫法检测3组血清孕酮及人绒毛膜促性腺激素(hCG)水平,采用Westernblot印迹杂交技术对3组流产绒毛组织中LIF的表达进行测定。结果难免流产组血清孕酮及hCG水平为(36.2±18.4)nmol?L及(13.6±3.1)kU?L显著低于正常早孕组(93.6±27.3)nmol?L及(70.2±15.9)kU?L和先兆流产组(89.5±25.2)nmol?L及(60.3±13.7)kU?L,差别均有显著性意义(均P<0.01),先兆流产组与正常早孕组间差别无显著性意义(均P>0.05);正常早孕组、先兆流产组和难免流产组绒毛组织中LIF平均相对表达量依次降低,分别为1.12±0.11、1.04±0.08、0.23±0.02,难免流产组LIF表达量显著低于其它两组(P<0.01),先兆流产组与正常早孕组间LIF的表达差别无显著性意义(P>0.05)。结论LIF在早孕者流产绒毛组织中的表达量降低,可能与hCG及孕酮分泌下降有关,是最终导致难免流产的原因之一。 Objective To investigate the expression of leukemia inhibitory factor LIF in the chorionic villi and its relation to spontaneous abortion. Method Pregnant women accepting induced abortion voluntarily were divided into 3 groups:normal early pregnancy group threatened abortion group and inevitable abortion group with 30 cases in each group .The serum and chorionic villi samples were collected. The expression of LIF in the chorionic villi was detected by Western blot and the serum levels of progesterone human chorionic gonadotropin hCG were measured by radioimmunoassay. Results Serum levels of progesterone and hCG in inevitable abortion group(36.2±18.4)nmol L and (13.6±3.1)ku L were decreased significantly P<0.01 compared with those of normal early pregnant group(93.6±27.3)nmol L and(70.2±15.9)kU Land threatened abortion group(89.5±25.2)nmol L and (60.3±13.7)kU L.There were no significant differences in serum levels of progesterone and hCG between normal early pregnancy group and threatened abortion group P>0.05. The expression level of LIF in inevitable abortion group 0.23±0.02 was lower significantly than that in normal early pregnancy group1.12±0.11 and threatened abortion group1.04±0.08 P<0.01. There was no statistically significant difference in the expression of LIF between the normal early pregnancy group and threatened abortion groupP>0.05. Conclusion The reduction of LIF expression in the chorionic villi of early pregnancy may decrease the serum progesterone and hCG levels and cause inevitable abortion.
