摘要
用体外细胞液体培养法研究组胺H_2受体激动剂4-甲基组胺(4-MH)、H_2受体拮抗剂西咪替丁和钙调素拮抗剂三氟啦嗪(TFP)对小鼠粒单白血病WEHI_3细胞增殖的影响。结果表明:4-MH呈剂量依赖性抑制WEHI_3细胞增殖。10 ̄(-4)mol/L西咪替丁可阻断这种效应。10 ̄(-5)─10 ̄(-4)mol/LTFP阻断WEHI_3细胞增殖,10 ̄(-6)mol/LTFP对细胞影响不大,但与高浓度4-MH合用有协同抑制效应。
The effects of
4-MH(4-methylhistamine,an agonist of histamine
H_2-receptor),cimetidine(an antagonist of histamine H_2-receptor)and
TFP(trifluoperazine, a calmodulin antagonist)onthe proliferation of
WEHI_3 cells(a myelomonocytic leukemia cell line)were investigated in
theliquid culture in vitro.It was shown that 4-MH at concentrations
ranging from 10 ̄(-7)mol/L to 10 ̄(-5)mol/L significantly inhibited
the proliferation of WEHI_3 cells (P<0.05).These inhibitoryeffects
were displayed in a dose-dependant manner and were completely blocked
by the additionof cimetidine at concentration of 10 ̄(-4)mol/L,TFP
caused a dose-dependent inhibition of theproliferation of WEHI_3
cells at concentrations ranging from 10 ̄(-5)mol/L to 10 ̄(-4)mol/L,
but didnot affect the growth of WEHI_3 cells at the low
concentrations(<10 ̄(-5)mol/L).TFP at lowconcentration(10 ̄(-6)mol/L)
exhibited synergetic inhibitory effect with high concentrations
of4-MH(>10 ̄(-5)mol/L)on the proliferation of WEHI_3 cells. The above
findings suggested thatTFP and 4-MH could inhibit the proliferation
of WEHI_3 cell and 4-MH could influence theproliferation of WEHI_3
cell by stimulating histamine H_2 receptor. Calmodulin probably
wasinvolved in the mechanism of growth regulation of WEHI_3 cells by
4-MH.
