缺氧复合失血性休克动物血浆对血管内皮细胞与白细胞粘附的影响 被引量:1

摘要 本实验观察缺氧(模拟海拔4000m高原24h)复合失血性休克24h山羊血浆(SP)对培养的肺动脉内皮细胞(PAEC)与多形核白细胞(PMN)粘附的影响,并对其机制进行了探讨。结果表明,PAEC在含25%浓度8P的培养液中孵育10mh─12h后,与PMN粘附率明显增加(27.4%─46%,与对照组5%相比P<0.01);温育12h末PAEC与PMN的粘附力也比孵育3h者明显增加(P<0.01);血小板激活因子(PAF)拮抗剂BN52021对10─30min末PAEC─PMN粘附有一定的抑制作用(P<0.01),但对1─12h末的粘附无明显作用;膜稳定剂利多卡因能明显抑制PAEC─PMN早期(10min─3h)粘附,但对晚期(6─12h)粘附无明显作用。PAEC在含50%SP的培养液中孵育早期(1─3h末)细胞活力明显升高,6─12h后明显降低,24h后回升至对照水平;PMN在含SP的培养液中孵育1h未,细胞活力明显升高(P<0.01),升高的程度依赖于SP浓度(r=0.980,P<0.01)。上述结果提示:SP可激活PAEC和PMN,增加PAEC─PMN粘附;PAEC与PMN的激活及二者的粘附可能在缺氧复合失? The effects of plasma from goat under hypoxia and hemorrhagic shock onleukocyte-endothelial cell adhesion were studied. Goats(exposed to simulated 4000m high alti-tude in hypobaric chamber for 24h prior to hemorrhage)were bled to and maintained at a meanarterial pressute of 6.OkPa for 60min and then resuscitated.Plasma samples were collected at 24hafter resuscitation(shock plasma, SP).The adhesion of polymorphonuclear leukocytes(PMN)from rabbits to bovine pulmonary artery endothelial cells(PAEC)increased significantly whenPAEC were incubated in 25% SP for 10min 12h.The adhering force of PAEC-PMN also in-creased significantly after incubation with SP for 12.Platelet activating factor(PAF) inhibitorBN 52021 inhibited PAEC-PMN adhesion during 10min-30min.Lidocaine inhibited the earlyadhesion(10min-3h).Cell viability of PAEC incubated in medium containing 50%SP increasedduring the early period(1-3h),but decreased during the later period(6-12h),and recovered tocontrol value at 24h.Cell viability of PMN incubated in media containing SP increasedsignificantly and was dependent on SP concentration.It is concluded that SP can increasePAEC-PMN interactons by activating PAEC and PMN,which may in turn play an importantrole in the course of changes during hypoxia and hemorrhagic shock.
