摘要
采用LichrospherC18柱,以甲醇和水为流动相,流速0.3mL/min,甲醇从60%~100%线性梯度洗脱25min,液质联用,电喷雾(ESI),对己烷雌酚及其衍生物己烷雌酚单(双)丁酸乙酯基醚分别进行了分离和结构鉴定。以混合酸酐法将己烷雌酚单羧基丙基醚(HESMCPE)与载体蛋白偶联,形成完全抗原,并以紫外扫描分析完全抗原的偶联率。经计算,HESMCPEBSA偶联率为36。
Using liquid chromatography /spectrometry (ESI), phase B (methyl alcohol), phase A (water), 0.3 mL/min of flow rate, a linear gradient from 60% to 100% of phase B and eluate for 25 min on a Lichrospher C-18 column, hexoestrol and it’s derivates hexoestrol-mono-ether-butyrate-ethyl (HES-MEBE), hexoestrol-di-ether-butyrate-ethyl (HES-DEBE) were separated and identified. The conjugates of hexoestrol-mono-caroxyl-propyl-ethyl (HES-MCPE) with proteins as synthetic antigens were prepared by the mixed anhydride method and the conjugation rate was analyzed by UV scanning. The conjugation rate is 36.
