摘要 目的:利用体外培养探讨受精卵显微注射的影响因素的作用。方法:获取小鼠受精卵,在其他条件固定的情况下,分别比较不同的注射基因浓度,在CZB(加HEPES)操作液中显微操作时间以及注射前受精卵的质量对显微注射后的胚胎体外培养结果的影响。结果:基因浓度越高,受精卵发育越差,操作时间不同的卵发育无明显差异,质量好的受精卵发育较好,质量差的几乎不发育。结论:选用较低的基因浓度,良好的操作介质CZB(加HEPES),使用质量好的受精卵,会提高显微注射的效率。 Objective: To study factors affecting mouse zygotes microinjection in vitro. Methods: Mice zygotes were extracted. When the other conditions were stable, the effect of different concentration of the microinjected DNA, and different timing of micromanipulation in CZB(adding HEPES) medium, different quality of zygotes on microinjection was investigated in vitro. Results: The higher the concentration of the microinjected DNA, the worse the development of zygotes. There was no different in culture between different timing of micromanipulation. Zygotes with good quality grew better. Zygotes with bad quality almost cannot grow. Conclusion: Lower concentration of the microinjected DNA, choosing CZB(adding HEPES) and zygotes with good quality can enhance efficiency of microinjection.
