恶性疟原虫重组质粒pVXORFl-PfMSP2-HBS在小鼠骨骼肌的表达

摘要 目的观察恶性疟原虫海南分离株(FCC1/HN)裂殖子表面蛋白2(MSP2)融合HBsAg基因片断真核表达质粒pVXORFl-PfMSP2-HBS在小鼠骨骼肌的表达。方法KM小鼠分3组,用0.5%盐酸布比卡因对3组小鼠行预处理后,在同一部位Ⅰ组注射浓度为1μg/μl的重组质粒pVXORFl-PfMSP2-HBS100μl,Ⅱ组注射浓度为1μg/lμ的质粒pVXORFl-HBS100μl,Ⅲ组为正常对照组,注射pH7.4PBS100μl。分别在免疫后4周和8周用免疫组织化学实验检测小鼠注射部位肌肉组织中重组蛋白MSP2的表达。结果小鼠经DNA疫苗注免疫4周和8周后肌肉组织呈特异性阳性反应。结论重组质粒pVXORFl-PfMSP2-HBS免疫小鼠后,肌肉组织有重组蛋白MSP2的表达。 Objective To explore the recombinant protein expression status in muscle tissue of mice vaccinated with plasmid pVXORFl-PfMSP_2-HBS of Plasrnodium falciparurn FCCl/HN strain. Methods The recombinant plasmid pVXORFl-PfMSP_2-HBS, pVXORFl-HBS and pH7.4 PBS were injected into 3 groups of KM mice respectively by intramuscular route, and the expression product of DNA vaccine was detected with immunohistochemistry assay 4 weeks and 8 weeeks after immunization. Results The specific positive reaction of the immunohistochemistry assay were observed in muscle tissue in immune group when vaccinated with plasmid pVXORFl-PfMSP_2-HBS,while regatiue reaetion was obserred in the mice of other two groups. Conclusion After vaccinating KM mice with plasmid pVXORFl-PfMSP_2-HBS by intramuscular injection directly, the recombinant protein MSP2 could be expressed in muscle tissue of mice.
