120例人脑胶质瘤组织P73蛋白表达分析

摘要 目的研究P73蛋白在人脑胶质瘤中的表达,探讨其在胶质瘤发生、发展和恶性变中可能作用.方法应用免疫组化方法(ABC法)分别检测胶质瘤Ⅰ~Ⅱ级20例,Ⅲ、Ⅳ级各20例;另外随机选取星形细胞瘤、少枝胶质瘤、室管膜瘤和胶质母细胞瘤各15例;同时取正常脑组织11例作为对照组.结果 P73蛋白的表达在Ⅰ~Ⅱ级胶质瘤组和正常脑组织无显著性差异(X2=0.818,P>0.05),Ⅲ级胶质瘤组和正常脑组织有显著性差异(X2=5.011,P<0.05),Ⅳ级胶质瘤组和正常脑组织有显著性差异(X2=6.229,P<0.05),Ⅰ~Ⅱ级胶质瘤组和Ⅲ、Ⅳ级胶质瘤组比较有统计学意义(P<0.05),星形细胞瘤、少枝胶质瘤室和管膜瘤与正常脑组织组比较无统计学意义(P>0.05),胶质母细胞瘤与正常脑组织组比较有统计学意义(x2=4.547,双侧P=0.051,单侧P=0.040).结论①P73蛋白的表达在肿瘤的病理分级间有明显差异,随分化程度低而表达率增高,说明了P73基因可能参与胶质瘤的发生,发展和恶性变;②除胶质母细胞瘤外,其它细胞类型的胶质细胞瘤中的P73的表达均与正常脑组织无显著性差异;③人胶质瘤组织中可能是DNNp73表达占相对优势. Objective To explore the expression of P73 in human glioma and study the role of P73 in the developing and malignantly changing progress of human glioma. Methods 120 cases of human glioma, including 20 cases of grade Ⅰ~Ⅱ, 20 cases of grade Ⅲ, 20 cases of grade IV, 15 cases of astrocytoma, 15 cases of oligodendrocytoma, 15 cases of ependymoma and 15 cases of spongioblastoma were studied.11 cases normal celebral tissue were taken as control group. Immunohistochemistry (ABC)was used to determine the expression of P73 in gliomas and normal cerebral tissues. Results There was no significant difference in the expression of P73 in the grade Ⅰ~Ⅱ glioma and normal cerebral tissue (χ~2=0.818, P>0.05).There were significant difference between the expression of P73 in the grade Ⅲ and IVglioma and normal cerebral tissue (P<0.05).When tumor cell classification was concerned, only the expression of P73 in glioblastoma had a significant difference from the cerebral tissue. Conclusion ①The lower grade of gliomais, the higher the expression of P73 gene would be. The over expression of P73 gene in the malignant glioma,suggested that P73 gene should take part in the developing and malignant lychanging progress of human glioma. ②The expression of P73 in glioblastoma showed no significant difference as compared with the cerebral tissue. ③DNp73 expression is dominant in the expression of p73 in human glioma.
