糖尿病鼠心组织损伤机制及二甲双胍的保护作用 被引量:1

摘要 目的探讨糖尿病鼠心组织损伤的可能机制及二甲双胍的保护作用。方法腹腔注射链脲佐菌素建立动物模型,采用RT-PCR法测定各组实验鼠心组织RAGEmRNA的表达。结果糖尿病模型组实验鼠心组织RAGEmR-NA表达增加,其RAGE/β-actin相对值明显高于对照组(P<0·01)。应用二甲双胍治疗后,其心组织RAGEmRNA表达较模型组显著降低,其RAGE/β-actin相对值显著低于模型组(P<0·01)。结论RAGEmRNA的高表达可能参与了糖尿病鼠高血糖及糖基化终产物造成的心组织损伤,而二甲双胍对糖尿病鼠心组织损伤有一定的保护作用。 Objective To investigate the possible mechanism of heart tissue damage and protective effect of metformin in diabetic mice. Methods Animal model was induced by intra-abdominal injection of streptozotocin. The expression of (Receptor for Advanced Glycation End Products) RAGE_mRNA in mouse heart tissue was examined by RT-PCR. Results The RAGE_mRNA expression was increased in heart of experimental diabetic mice. RAGE/β-actin ratio was significantly higher than the control (P<0.01). After treatment with metformin, RAGE_mRNA expression decreased significantly and RAGE/β-actin ratio was obviously lower than the control. Conclusion The high expression of RAGE_mRNA may participate in the heart damage of diabetic mice. Metformin can protect the heart tissue from the impairment of hyperglycemia and advanced glycation end product.
