中国人TFPI-2基因的克隆及测序分析 被引量:5

摘要 目的对中国人的组织因子途径抑制物-2(TFPI-2)基因进行克隆并测序。方法从人新鲜肝组织提取总RNA,经逆转录PCR(RT-PCR)扩增出TFPI-2的全长cDNA,经亚克隆、筛选、鉴定后,进行正反测序。结果中国人TFPI-2基因cDNA全长1222bp,除258bp、585bp和884bp处与目前GenBank中收录的国外学者克隆的TFPI-2基因序列不同外,其他完全一致。其序列Gen-Bank登记号TFPI AY691946。结论中国人TFPI-2基因序列与已经报道的西方人的TFPI-2基因序列基本相同,但三处单核苷酸多态性的意义何在,目前尚不清楚。 Objective To clone and sequence the Chinese tissue factor pathway inhibitor-2(TFPI-2) gene (full length cDNA).Methods RNA was isolated from human liver and full length TFPI-2 DNA was amplified by RT-PCR. The cloned gene was inserted into plasmid and sequenced from forward and reverse direction.Results Sequencing results showed that the cloned Chinese TFPI-2 gene has three bases different from that registered in GenBank.The seuencing results has been transmitted and received by GenBank,accession number is TFPI AY691946.Conclusion The Chinese TFPI-2 gene sequence has three different bases with that of western and the role of the single nucleotide polymorphism need further study.
