摘要
转基因小麦B72-8-11b中外源品质基因1Dx5表达量是内源相应基因表达量的6倍。利用小麦转基因品系为父本,常规品种鄂麦12、鄂麦18和日喀则8号分别为母本,配置3个杂交组合。采用SDS-PAGE技术,检测各组合亲本、F1、F2代的HMW-GS组成,研究转基因小麦B72-8-11b中外源品质基因1Dx5表达的遗传。结果表明:外源1Dx5基因有功能拷贝整合在1个位点,如同内源品质基因,遵从孟德尔遗传模式。这对育种选择策略的制订具有指导意义,也表明了基因枪转化法能够使得外源基因有功能拷贝整合在1个位点并能稳定传递。
The transgenic gene 1Dx5 was over-expressed in the transgenic wheat line B72-8-11b, with the expression amount more five times than the corresponding native gene in non-transgenic elite lines of wheat. The parents, F1 and F2 generations from 3 crosses, with male parent being transgenic wheat, were tested for HMW-GS compositions by SDS-PAGE analysis, so as to explore the heredity of transgene 1Dx5 expressed in transgenic wheat line B72-8-11b, generated by particle bombardment. The results showed that the functional copies of transgene 1Dx5 were integrated in one locus, as a dominant gene, abiding by Mendelian genetic law. It will be beneficial to deciding the selection strategy in breeding program, showing the stable heritability of single gene locos into which transgenic gene functional copies were integrated by particle bombardment transformation.
