不同来源莪术中莪术醇、吉马酮、莪术二酮GC-MS定量分析 被引量:20

Quantitative Determination of Curcumol, Germacrone and Curdione from Zedoray Rhizome by GC-MS

摘要 目的:测定并比较不同来源莪术中莪术醇、吉马酮、莪术二酮的含量。方法:采用甲醇超声提取,GC-MS选择离子监测法测定3种成分含量。色谱条件:HP-5MS毛细管柱(30 m×0.25 mm×0.25μm);柱流速为1 mL·min-1,柱温:100℃(2 汽化室温度250℃;载气为高纯He(99.999%)。质谱条件:EI离子源,电子能量70 eV;四极杆温度150℃;EM电压2165 V;接口温度280℃;溶剂延迟 3 min;m/z范围40-550 amu。结果:莪术醇、吉马酮和莪术二酮分别在12.4-124.0,12.4-82.3,13.0-86.7 ng线性关系良好;r值分别为0.9990,0.9981,0.9989。不同来源莪术中莪术醇含量均极低,大多数样品均未检出;所有样品中均可检出吉马酮,但含量差异较大(5.479 mg·g-1vs0.459 mg·g-1);莪术二酮在部分样品中未检出。结论:不同来源莪术中莪术醇、吉马酮和莪术二酮的含量差异显著。 Objective:To determine and compare the contents of curcumol, germacrone and curdione in Zedoray rhizome from different species and places. Methods:Ultrasonication with methanol was used for sample preparation, and quantitative determination of analytes was performed by GC - MS. GC conditions; HP-5MS capillary column (30 m×0.25 mm×0.25μm) ,flow rate was 1 mL·min-1,column temperature program:100℃(2min) injector temperature: 250℃,Gas; He (99.999% ). MS conditions; El ionization mode,electron energy 70 eV ,quadropole temperature: 150℃,EM voltage 2165 V,interface temperature:280℃,solvent delay for 3 min,m/z:40-550 amu. Results: The linear ranges of curcumol,germacrone and curdione were 12.4-124.0,12.4-82.3,13.0-86.7 ng with coefficient values (r) 0.9990,0.9981 and 0.9989,respectively. The content of curcumol in Zedoray rhizome was very low, and even was not detected in most samples; germacrone was detected in all the samples, but the difference of the content in different samples was obvious (5.479 mg·g-1vs 0. 459 mg·g-1) ; curdione was under the limit of quantitative determination in some samples. Conclusion:The contents of curcumol, germacrone and curdione in Zedoray rhizome from different species and places were significantly different.
