摘要 以人血清白蛋白作基质制备包含化疗药丝裂霉素C的磁性药物微球,并考察其体内分布及体外释药,动物安全性及药效。结果表明,该微球能够浓集滞留于靶位并缓慢释放药物。动物实验证明其安全可靠,疗效比单独使用阿霉素提高一倍多。这表明,磁性微球有希望作为抗肿瘤药物载体应用于临床靶向给药系统以取得局部化疗和物理栓塞的综合治疗效果。 Magnetic mitomycin C-bearing microspheres 20-120μm in diameter were prepared with human serum albumin as a matrix.The distribution in vivo and releasing behavior of drug in vitro,toxic response and pharmacodynamics of the microspheres were studied.The external magnetic field was designed and its characteristics were tested.The results showed that the microspheres could be held up in the target region,sustainedly release the drug(released amount 70%/24hrs)and the drug condensity inside the target could be greatly increased to 10-150 times.in animal injection of the microspheres into the body not only had not caused any toxic reactions but therapeutic effect was also enchanced as compared with merely drug administration(ADM).It is suggested that the microspheres can be hopefully applied clinically.
