摘要
采取不同破壁方法对玉米花粉进行破壁处理,并利用正交设计对玉米花粉多糖的分离提取条件进行研究。结果表明,酶解破壁法和机械破壁法均显著优于温差破壁法,而酶解破壁法与机械破壁法差异不明显。机械破壁法操作方便且成本较低,因此认为适合在多糖提取中使用。花粉破壁处理是影响多糖产量的主要因素,其次为料液比、浸提温度和浸提时间。玉米花粉多糖分离提取的最优条件为:采取机械破壁法,料液比为1:6,浸提温度为70℃,浸提时间2h。
Corn pollen was processed by different wall-breaking methods before extraction. The orthogonal test design was used in extraction optimization of pollen polysaccharide of corn (PPC). It showed that the enzyme wall-breaking method and mechanical wall-breaking method were significantly better than temperature -difference wall-breaking method, but there were no significant differences between the enzyme wall -breaking and mechanical wall -breaking method. As mechanical wall breaking had advantages of convenience and low cost over enzyme wall -breaking method, it was recommended in PPC extraction. The pollen wall-breaking process was the main factor influencing PPC extraction, then the proportion of pollen to solution, the extraction temperature and extraction time. The optimized conditions for PPC were mechanical wall-breaking method, 1:6 proportion of pollen to solution, 70℃ of extraction temperature for 2 hours.
