盐酸去氧肾上腺素对体外培养兔角膜内皮细胞的毒性作用 被引量:3

摘要 目的观察盐酸去氧肾上腺素对体外培养家兔角膜内皮的作用,探讨临床应用去氧肾上腺素的可能毒性作用。方法体外培养家兔角膜内皮细胞,传代后加入0.625mg/ml、1.25 mg/ml、2.5 mg/ml、5mg/ml 4组不同浓度盐酸去氧肾上腺素,观察光镜下细胞的形态变化和增殖情况,72h 后 MTT 法检测各组细胞的吸光度值,对比不同浓度实验组细胞吸光度并进行统计学处理。结果加药4h 后5mg/ml 实验组细胞出现空泡;30h 后2.5mg/ml、5mg/ml 实验组细胞部分死亡;60h 后1.25mg/ml、2.5mg/ml 实验组原空泡变细胞大部分死亡;0.625mg/ml 实验组细胞无明显变化。1.25mg/ml、2.5mg/ml、5mg/ml 实验组细胞吸光度值与空白及阴性对照的差异经统计学分析有极显著意义(P<0.01);4个阴性对照组分别与空白对照组相比较,差异无统计学意义(P>0.05)。结论盐酸去氧肾上腺素对体外培养兔角膜内皮细胞有损伤作用,并有时间和浓度相关性。 Purpose To observe the toxicity of Phenyleprine Hydrochloride on cultured rabbit corneal endothelial cells and to explore its safety for clinical use.Methods Rabbit corneal endothelial cells in culture were treated with different concentration of Phenylephrine(0.625 mg/ml,1.25 mg/ml,2.5 mg/ml and 5 mg/ml),cellular changes in cell morphology were observed under phase-contrast microscope in different time point,and MTT assay on 414 nm was used to detect the proliferation of the cells.Results Phenyleprine caused vacuolar degeneration of cells from 4h to 60h and most of the cells died eventaally.Toxic effect on cells was further confirmed by MTT assay(P<0.01)at 1.25 mg/ml,2.5 mg/ml,5 mg/ ml.There was no toxicity effect on corneal endothelial cells at 0.625 mg/ml(P>0.05).Conclusions The toxicity of Phenylephrine to rabbit corneal endothelial cells is associated with its concentration and duration of exposure.
