病毒感染小鼠心肌中内皮素及其A受体表达 被引量:1

Expression of endothelin and its receptor A in the myocardium of infected mice

摘要 目的:探讨病毒性心肌炎小鼠心肌组织中是否存在有血管收缩作用的内皮素及内皮素A受体mRNA。方法:实验于2003-10/2004-04在哈尔滨医科大学克山病研究所完成。选用雄性清洁级Balb/c小鼠40只,腹腔注射嗜心肌柯萨奇B组病毒3型制备病毒性心肌炎模型,用反转录-聚合酶链反应技术半定量检测病毒感染后即刻,9,14及21d小鼠心肌中的内皮素A受体-mRNA表达。用免疫组化方法测定病毒感染后即刻及感染后14d心肌中内皮素表达情况。结果:①病毒感染后14d心肌中内皮素A受体mRNA表达明显高于病毒感染后即刻(0.672±0.0828,0.501±0.085,q=4.888,P<0.05)。病毒感染后7,21d心肌中内皮素A受体mRNA的表达高于感染后即刻,但无明显差异(q=1.471,3.089,P>0.05)。②病毒感染14d后心肌中内皮素表达明显高于病毒感染后即刻(0.034477±0.017676,0.00288±0.001768,t=4.3568,P<0.05)。结论:内皮素系统参与病毒性心肌炎的损伤过程。 AIM:To investigate whether there is endothelin, which has the role of vasoconstriction, and endothelin receptor A mRNA in the myocardial tissue of mice with viral myocarditis. METHODS:The experiment was carried out in the Institute of Ke-shan Disease, Harbin Medical University between October 2003 and April 2004. Forty male Balb\c mice of clean grade, were made into models of viral my-ocarditis by intraperitoneal injection of myocardium-like Coxsackie virus B types 3. The expression of endothelin type A re-ceptor-mRNA in the myo-cardium of mice was detected semi-quantita-tively with reverse transcrip-tion-polymerase chain reaction immediately and 9, 14 and 21 days after vi-ral infection. The expression of en-dothelin in myocardium was detected with immunohistochemistry imme-diately and 14 days after infection. RESULT:① The expression of endothelin type A receptor-mRNA 14 days after viral infection was obviously higher than that immediately after viral infection (0.672±0.082 8, 0.501±0.085, q=4.888, P< 0.05); It was higher 7 and 21 days after viral infection than imme-diately after viral infection but not obviously (q=1.471, 3.089, P > 0.05). ② The ex-pression of endothelin in myocardium 14 days after viral infection was sig-nificantly higher than that immediately after viral infection(0.034 477 ±0.017 676, 0.002 88±0.001 768, t=4.356 8, P < 0.05). CONCLUSION:The endothelin system is involved in the injured process of virus myocarditis.
