芪丹通脉片对动脉粥样硬化大鼠主动脉抗原CD40和胞间黏附分子1mRNA表达的影响 被引量:7

摘要 目的:探讨芪丹通脉片对动脉粥样硬化大鼠主动脉抗原CD40、胞间黏附分子1的影响。方法:实验于2003-04/08在解放军第四军医大学实验动物中心、西京医院检验科分生实验中心完成。将72只SD雄性大鼠随机平均分为6个实验组:阴性对照组(n=12)给以普通饮食,模型组(n=12)给予高脂、高胆固醇饮食,其余4组在给予高脂、高胆固醇饮食建立动脉粥样硬化大鼠模型同时分为芪丹通脉片低剂量组[0.36g/(kg·d)]、中剂量组[1.08g/(kg·d)]、高剂量组[3.24g/(kg·d)]和阳性对照辛伐他汀组[4mg/(kg·d)]。14周后通过反转录多聚酶链反应,检测大鼠主动脉上CD40、胞间黏附分子1表达。结果:模型组表达CD40、胞间黏附分子1分别为1.730±0.022,1.55±0.17,均明显高于阴性对照组(0.390±0.012,0.43±0.09)(P<0.05);给予芪丹通脉片大鼠CD40、胞间黏附分子1表达显著降低,高浓度组(0.555±0.020,0.52±0.11)结果接近辛伐他汀组(0.400±0.016,0.46±0.12)。结论:动脉粥样硬化大鼠主动脉CD40、胞间黏附分子1表达增强,使用芪丹通脉片可降低CD40、胞间黏附分子1表达,且其低、中、高浓度组表达依次减少。 AIM: To study the effect of Qidantongmai tablet(QDTMT) on CD40 and intercellular adhension molecule-1(ICAM-1)expression of aorta in rat with atherosclerosis (AS). METHODS: The experiment was finished in Center of Animal Laboratory and Department of Clinical Laboratory of Fourth Military Medical University of Chinese PLA from April to August 2003.Totally 72 male SD rats were divided into six experimental groups. Control group was assigned randomly to receive normal diet (n=12).Model group was given high fat and cholesterol diet to make AS models (n=12).The other four study groups were given high fat and cholesterol diet to make AS models.,and were divided into low dose QDTMT group[0.36 g/(kg perday)], middle dose QDTMT group[1.08 g/(kg perday))],high dose QDTMT group[3.24 g/(kg perday))]and Simvastatin group[4 mg/(kg·d)]for 14 weeks. Then the expression of CD40,ICAM-1 in aorta in each group was detected by reverse transcript polymerse chain reaction(RT-PCR). RESULTS: The expression level of CD40 and ICAM-1 in model group (CD40: 1.730±0.022, ICAM-1: 1.55±0.17) was eobviously higher than that in control group(CD40: 0.390±0.012, ICAM-1: 0.43±0.09) (P< 0.05). The significant decrease of CD40 and ICAM-1 mRNA ex-pression was detected in groups given QDTMT. The result of higher dose group (CD40: 0.555 ±0.020, ICAM-1: 0.52±0.11) was close to the Simvastatin group (CD40: 0.400±0.016, ICAM-1: 0.46±0.12). CONCLUSION: The expression of CD40 and ICAM-1 in aorta in rat with atherosclerosis is increased.QDTMT can reduce the express of CD40 and I-CAM-1.This tendency dependeds on dose. The expression of the low, mid-dle and high dose QDTMT groups reduces in order.
