大鼠上矢状窦后1/3夹闭对其回流区组织的影响──自由基、脑水肿及病理改变 被引量:4

The Cerebral Edema,Superoxide Dismutase and Histological Changes after Posterior 1/3 Occlusion of Sagittal Sinus in Rats

摘要 外伤后上矢状窦闭塞的死亡率很高.通过大鼠上矢状窦后1/3夹闭实验,观察了其回流区域皮层组织脑水含量、组织超氧化物歧化酶(SOD)和病理学的改变,探讨其回流区域所受影响.结果提示,夹闭4小时后开始出现脑水肿,8~12小时达到高峰,以后逐渐减轻.结扎后矢状窦旁皮层组织SOD含量明显低于颞叶皮层组织.光镜下24小时窦旁皮层神经元细胞出现核固缩.电镜观察,2小时窦旁皮层神经细胞线粒体肿胀,4小时线粒体脊破坏.由此表明,矢状窦后1/3的急性梗阻导致的损害局限于窦旁皮层组织,不可逆损害发生于结扎后2~4小时. We observed the cerebral edema,superoxide dismutase (SOD) and histological changes to investigate the effluence of the drainage area after acute occlusion of posterior 1/3 sagittal sinus.The changes of brain water weight wet versus dry were measured at 4,8,12 and 24 hours ( n =5) after posterior 1/3 sagittal sinus ligation.The SOD of paresagittal and temporal tissues was measured at 8 hour ( n =8) after the occlusion.The paresagittal tissues were removed at 2,4,8,12,24 hours ( n =4) and prepared for histological examination.The changes of water weight were 78.7±0.8 in the 4 hour group,82.9±0.6 in the 8 hour group,80.5±0.4 in the 12 hour group,79.4±4.6 in the 24 hour group.Compared with the control group (78.2±0.4),there was significant difference except the 4 hour group.The content of SOD was 4.00±0.41 nU/mg in paresagittal tissues and significantly lower than that of the temporal tissues (4.56±0.52 nU/mg).The histological damage was limited to the paresagittal areas under the high microscope.The electron microscopic changes included swelling of the mitochondria after 2 hours,loss of cristae after 4 hours and phyknotic nucleus after 24 hours.These indicated that cerebral edema appared 4 hours after posterior sagittal sinus ligation,the tissue damage was limited to the paresagittal areas,and the irreversible damage appeared after 2 hours.(Original article on page 219)
