摘要
目的:以腺病毒为载体,研究外源性突变型p27(p27mt)基因对大肠癌的体内抑制作用.方法:应用细胞移植法把人大肠癌SW480细胞接种于BALB/c裸鼠皮下,构建大肠癌裸鼠模型,将已成功构建的腺病毒介导的突变型p27(Adp27mt)及LacZ采用瘤体内直接注射法注射到瘤体内,与对照组比较,绘制肿瘤生长曲线,计算肿瘤生长抑制率,并进行细胞凋亡的相关检测.结果:Adp27mt基因治疗组肿瘤生长明显受到抑制,有明显的促凋亡作用,与LacZ、对照组比较,有显著性差异(P<0.01).结论:Adp27mt基因对大肠癌裸鼠模型的体内肿瘤有明显的抑制作用.
AIM: To investigate the inhibitory effect of extrinsic p27mt gene mediated by adenovirus vector on colorectal carcinoma nude mice. METHODS: The model of colorectal carcinoma nude mice was established by inoculating the SW480 cells of human colorectal carcinoma. The Ad-p27mt and Lac-Z was injected into the tumors of treatment groups respectively. The growth curves and the growth inhibition rates of tumors in the treatment groups were analyzed and compared with those in the control group. RESULTS: The Ad-p27mt obviously suqpressed the growth of tumors and promoted apoptosis of SW480 cells, which were significantly different compared with those in the control group (P<0.01). CONCLUSION: The Ad-p27mt has obvious inhibitory effect on the tumor growth of colorectal carcinoma in nude mice in vivo.
