摘要
目的探讨p53蛋白在冷冻联合5-FU诱导G422胶质瘤细胞凋亡过程中表达的改变,欲为冻化疗联合应用治疗胶质瘤提供理论依据。方法建立G422胶质瘤动物模型,随机分为对照组、冷冻组、化疗组和冻化疗组;免疫组化法检测p53蛋白的表达,TUNEL法观察各组细胞凋亡的变化。结果冷冻与5-FU均能诱导G422胶质瘤细胞发生凋亡,两者的联合应用使肿瘤细胞凋亡率明显提高;p53蛋白在冷冻后6、12、24、48和72h阳性表达率分别为(19.9±4.1)%、(21.0±6.5)%、(22.7±6.6)%、(35.6±6.6)%和(31.8±6.1)%,其表达较对照组明显增加,尤以冻后48h最为明显,P<0.01。结论冷冻与5-FU联合应用能明显提高G422胶质瘤细胞凋亡率,冷冻后p53蛋白表达的增加在此过程中起着重要作用。
OBJECTIVE:To investigate the variation of p53 protein expression in apoptosis process of G422 glioma induced by cryotherapy and 5-fluorouracial(5-FU) and to raise a theoretical basis for clinical practice of cryochemotherapy on glioma. METHODS: A mouse experimental model with G422 glioma was established and the experimental mice were divided into four groups randomly:control group,chemotherapy group,cryotherapy group and cryochemotherapy group.TUNEL was used to detect glioma cell apoptosis of each group at different time points. The expression of p53 protein was analysed by immunohistochemical staining.RESULTS:Both cryotherapy and 5-FU induced apoptosis of G422 glioma, while the combination of them did a remarkably higher ration of it. The expressions of p53 protein at 6, 12, 24, 48, 72 h after cryotherapy ware (19.9±4.1)%, (21.0±6.5)%, (22.7±6.6)%, (35.6±6.6)% and (31.8±6.1)% respectively,which were more obvious than that of the control group, especially at 48 h, P<0.01. CONCLUSION: Combination of cryotherapy with 5-FU could enhance the apoptosis of G422 glioma, and variation of p53 protein expression may play an important role in the process.
