摘要
本研究目的是探讨四氧嘧啶糖尿病大鼠骨骼在骨量、超微结构和生物力学方面的变化,井观察胰岛素对这些病变的治疗作用。将50只雄性SD大鼠分成正常组、糖尿病组和胰岛素治疗组,3个月后处死,运用SPA法和QCT法测定股骨干及骺端骨量,电镜观察股骨及腰椎结构,并对股骨及腰椎进行生物力学测试。结果:四氧嘧啶糖尿病鼠3个月后骨骼出现了明显的病变:骨量明显降低、形态结构疏松、成骨功能丧失、骨质结构和材料性能也显著改变。胰岛素可纠正绝大多数糖尿病骨骼病变,但出现了骨骼脆性增加的副作用,望引起临床医师重视。
Abstract For the investigation of the pathological changesof bone densitv,ultrastructure and biomechanics onbone in alloxan diabetic rats,50 male SD rats weredivided into three groups of the normal,control,diabetic and diabetic treated with insulin。All animalswere sacrificed after 3 months and SPA and QCT wereused for the determination of bone density on femur。The ultrastructure of femur and lumbar spine wereobserved by electron microscope and the biomechanicswere tested.Thresults shOwedthat,alloxan diabeticrats suffered from significant diabetic osteopathy,bone density loss,and structural osteoporosis,deficient function of osteoblast and markeddeterioration of biomechanics on bone。Theadministration of insulin corrected most of thepathological changes,however,the vertebrae werestill more brittle. The authors reminded the clinicalphysicians paying attention to diabetic osteopathy thatwill certainly occur in human subjects。
