摘要
应用单细胞凝胶电泳检测技术(SingleCellGelAssay,SCG)对某钢厂不同生产车间工人外周血淋巴细胞单链DNA损伤进行检测。结果表明,CO组、CO+高温组工人的DNA损伤发生率高于高温组、粉尘组和对照组4~6倍,其DNA损伤的程度亦较严重(以接触CO者为最严重)(P<0.01),而高温组、粉尘组和对照组之间差异无显著性(P>0.05),提示外周血淋巴细胞单链DNA损伤率有可能用作职业有害因素对健康影响的早期监测指标。
he single cell gell( SCG )assay (namely,comet
assay)is a sensitive electrophoretic technique for detecting the presence of DNA single strand
breaks and alkali-labile damage in individual cell.The technique was used to evaluate the
levels of DNA darmage in peripheral blood lymphocytes of the workers exposed to
heat,CO,CO+heat and dust.The subjects in control group were the personnel with no
occupational exposure history.The percentage of DNA damage in CO,CO+heat were 4~6 times
higher than those in heat,dust and control.The darnaged levels were more severe too. The
previous data of the worker’s health indicated that cancer incidence of the CO group and
CO+heat group was 7 times higher than that of the control.t is suggested that the rate of DNA
damage in lymphocytes may be an early health monitoririg index for workers in some
occupational hazards.
