农杆菌介导的枳壳转基因条件优化途径建立研究 被引量:1

摘要 本文以枳壳上胚轴为对象,进行了农杆菌介导的抗菌肽shivaA基因转化影响因素研究,得到了转基因植株。研究结果表明:枳壳上胚轴的卡那霉素选择浓度为60μg/ml;采用头孢噻肟钠为农杆菌抑制剂较好。共培养基3天,共培养基中补充As100μM、0.5%脯氨酸对转化率的促进作用最大;延迟选择不利于转化率的提高。 s Transformation and regeneration of Poncirus trifoliata Raf. plant were studied by Agrobacterium-mediated. Transgenic plants with shivaA gene have been obtained. As a selection agent, the concentration of kanmaycin is 60 mg/L. The transformation frequeney of explants was promoted when 100 μM As & 0.5% proline were added in co-culture medium. It is no useful for anti-Km shoot producing when selectable cultivation is postponed. Foreign gene is proved going to P. trifoliata by PCR examine.
