摘要
目的观察子宫腺肌病灶组织细胞对米非司酮作用的敏感性。方法采用噻唑兰法观察米非司酮对22例子宫腺肌病患者腺肌病灶组织原代培养细胞体外生长状态的影响。实验分为1.25mg/L、2.5mg/L、5.0mg/L,3个药物浓度组,培养52h,测定OD值,并与空白对照组对比。结果1.25mg/L、2.5mg/L、5.0mg/L组及对照组的OD值分别为(0.219±0.044)(、0.185±0.038)(、0.152±0.033)、(0.262±0.068),5.0与1.25mg/L组有显著差异(P<0.01),5.0mg/L、2.5mg/L组与对照组有显著差异(P<0.01)。细胞生长抑制率2.5mg/L组(IR=29.5%)是1.25mg/L组(IR=16.6%)的1.79倍,5.0mg/L组(IR=42.0%)是2.5mg/L组的1.43倍。结论米非司酮抑制子宫腺肌病腺肌组织细胞的生长能力与药物浓度呈正相关。
Objective To observe the effects of mifepristone on the cultured adenomyotic cells. Methods With MTT assay,the growth state of premary cell culture trom adenomyotic implants of 11 women with adenomyosis was studied.The cells of each case were deveded into foru groups,three treat ment groups(1.25mg/L,2.5mg/L,5.0mg/L mifepristone,respectively) and one control group.cultured for 52 hours before the OD value was determined.Results The OD values of 1.25mg/L,2.5mg/L,5.0mg/L mifepristone and control group were (0.219±0.044),0.185±0.038) and (0.262±0.068) respectively.The OD value of 5.0mg/L groups had significantly lower OD values than the control group(P<0.01),and both 5.0mg/L and 2.5mg/L groups had segnificantly lower OD values than the control group(P<0.01).The inhibition rate of cell growth in 2.5mg/L group (IR=29.4%) was 1.79 times as much as that in 1.25mg/L,group(IR=16.4%),while in 5.0mg/L group (IR=42.0%) it was 1.43 times as much as that in 2.5mg/L group.Conclusion Mifepristone could suppress the growth of adenomyotic cells in vitro in a dose dependent manner.
