人参总皂甙对大鼠脑缺血再灌注后海马CA1区GABA表达及细胞凋亡的影响 被引量:1

摘要 目的探讨人参总皂甙对大鼠脑缺血再灌注损伤的保护作用及其作用机制。方法将40只Wis-ter大鼠随机分为正常对照组、假手术组、缺血再灌注组、治疗组1、治疗组2,采用线拴法制备大鼠脑缺血再灌注模型,在72h断头取脑,石蜡切片行GABA、TUNEL染色,计算凋亡细胞数及GABA阳性反应数。结果治疗组与缺血再灌注组比较海马CA1区凋亡细胞数减少,γ-氨基丁酸(GABA)阳性反应数增多(P<0.01)。结论人参总皂甙能明显增强局灶性脑缺血再灌注大鼠海马神经元GABA的表达,抑制神经细胞凋亡,而发挥神经保护作用。 Objective To study the protective effect and the functional mechanism of ginseng total sapomin on cerebral ischemia reperfusion injury in rats. Methods 40 adult Wister rats were randomly divided into 5 groups: normal group, sham-operated group, cerebral ischemia reperfusion group, treated group 1 and treated group 2. The model of focal cerebra ischemia reperfusion was made by string-embolic method. After 72 hours, the brains were removed and sliced up for GABA, TUNEL straining. The data of apoptosis cells and GABA expressive cells were calculated. Results Compared with cerebral ischemia reperfusion group, the treated groups had more apoptosis cells and fewer GABA expressive cells. Conclusions Ginseng total saponin may increase the expression of GABA in the hippocampus neurons of focal cerebral ischemia reperfusion rats, and play an important role in protecting neurons by inducing apoptosis.
