摘要
转化生长因子β1(TGF-β1)对两种不同恶性表型的膀胱移行上皮癌具有不同的调节作用。DNA合成抑制试验表明,5ng/mlTGF-β1可控制分化较好的647V细胞系[3H]-Thymidine的结合达60%;但在同样浓度下不能抑制反而刺激具有转移能力的253J细胞系的DNA合成。Northern杂交分析显示,FN与PAⅠ-1的表达对647V无明显影响。结果揭示TGF-β1对有或无转移能力的肿瘤细胞的不同调节作用可能影响肿瘤的复发与转移。
The effects of TGF-β1 on two transitional carcinorna cell lines were investigated.In a growth inhibition assay determined by 3H-thymidine incorporation, 647V cell line was about 40 ̄60% inhibited by TGF-β1( 5ng/ml), whereas,253J cell line was not responsive to the inhibitory effect of TGF-β1. By using northern analysis,we assayed gene response of both cell lines to TGF-β1 in term of FN and PAI-1 expression. Both of FN and PAI-1 genes were upregulated in 253J cell line.but slight changes were found in 647V. Thus 253J which was resistant to the growth inhibitory effect of TGF-B1 could response to TGF-β1 by extracellular matrix expression. However,647V which had slight changes of FN and PAI-1 expression was growth inhibited by TGF-B1.Our results suggested that different response to TGF-B1 might be an important distinction between metastatic and nonmetastatic tumor cells and may play an especially important role in tumor recurrence and metastasis.
