摘要
目的研究四氯化碳(CCl4)对大鼠胚胎脊髓神经元生长发育的毒性作用,并探讨其中的机制.方法采用大鼠胚胎脊髓神经元体外培养技术,研究不同浓度的CCl4(0.01~100.0 mg/L)对细胞分化和增殖的影响,并利用图像分析细胞形态的变化、硫代巴比妥酸测定丙二醛(MDA)和溴化四氮唑噻蓝分析蛋白总量.结果10.0~100.0 mg/L CCl4组随浓度的增加细胞数和集落形成率明显下降,细胞内MDA含量增加,蛋白质相对含量降低,并显示剂量效应关系.结论CCl4对体外培养的大鼠胚胎脊髓神经元分化和增殖的抑制作用,可能与其诱导蛋白质合成和脂质过氧化变化有关.
ObjectiveTo study the development toxic effect of carbon tetrachloride (CCl_4) on the embryonic spinal cord neurons of rats, and to investigate its mechanism. MethodsThe effects of different contents of CCl_4 (0.01~100.0 mg/L) on embryonic spinal cord cells of rats in vitro culture were observed. The changes of cell shapes were analyzed with image analysis. The content of MDA was determined with the method of TBA, and the total content of protein was determined with MTT. ResultsThe decreases of cell number and colonies were shown in all cells treated by CCl_4. The contents of protein were concentration-dependently decreased, and the levels of MDA were increased with the increasing dosage of CCl_4. ConclusionThe differentiation and proliferation inhibition of the rat embryonic spinal cord nerve cells in vitro culture might be associated with the changes of protein synthetics and lipid peroxidation induced by CCl_4.
