To investigate the effects of ligustrazine (LT) on hypoxic pulmonary vasoconstriction (HPV) in vivo, the lungs of nine male rates served as the experimental subjects. The status of pulmonary vasoconstriction was indicated by the levels of the perfused pressure (PP), under which the pulmonary blood flow was pumped at a constant rate. With the perfused lungs being ventilated with a mixture of 3 % oxygen, 4 % carbon dioxide and 93 % nitrogen, the level of PP increased from 2. 44 0. 24 to 3. 69 0. 54kpa,indicating that HPV was developed. Afterwards,LT at a dose of 4mg. kg-1 was administrated to the perfused pulmonary blood, and two hours later when PP returned to the primary high level, LT at a dose of 8mg. kg-1 was given subseqently. The PP was recorded continuously with a pressure monitor during the entire procedures. The results revealed that following the administration of LT 4mg. kg-1 or 8mg. kg-1,the level of PP decreased by 63% or 96% respectively. It is concluded that LT can inhibit HPV effectively, and the degree of the effect may be releted to the dosage.
