猪苓多糖、乙型肝炎疫苗和卡介苗对外周血单个核细胞免疫活性的影响

THE EFFECTS OF ZHULING-DUOTANG HEPATTTIS B VACCINE AND BACILLUS CALMETTE-GURIN ON IMMUNOACTIVITIES OF PERIPHERAL BLOOD MONONUCLEAR CELLS:AN IN VITRO RESEARCH

猪苓多糖联合乙型肝炎疫苗和潘生丁联合卡介苗用于治疗慢性乙型肝炎，经观察认为它们在抑制乙型肝炎病毒复制方面有较好的疗效；但其作用机理不清。本实验试图通过观察上述药物对正常人外周血单个核细胞（ＰＢＭＣ）免疫杀伤活性的影响，探索其发挥作用的机理。结果表明：猪芬多糖和卡介苗体外可增强正常人ＰＢＭＣ中免疫细胞的杀伤活性，提高ＰＢＭＣ细胞膜上白细胞介素之受体的表达并促进其分泌白细胞介素－２。因此认为上述作用是它们发挥疗效的机理之一。乙型肝炎疫苗体外无增强正常人ＰＭＢＣ中免疫细胞杀伤活性的作用；它联合猪芬多糖诱导正常人ＰＭＢＣ也不比猪芬多糖单独诱导出的杀伤活性高。

癦huling-duotang(a polysaccharide preparation of the Chinese traditional herb medicine polyporusum bellatus ) and hepatitis B vaccine(HB vaccine )”and “persantin and bacillus cain1eteguerin ( BCG)”have been used to treat chronic hepatitis B. To elucidate their therapeutic mechanism,we studied the effects of zhuling-duotang,HB vaccine and BCG on immunoactivities of nomal human peripheral blood mononuclear cells(PBMCs). Cytotoxicity of PBMCs incubated with such drugs against target cells K562, HepG_2 or2.2. 15 was detected by using ￣3H1dR release assay. IL-2 and IFN-r activities of the supernatant were assayed.Immunofluorescent analysis of molecular CD_4, CD_8, CD_16, IL-2R on the surfase of PBMCs weredone. The results showed that :(1) Zhuling-duotang and BCG could significantly increase the cytotoxicity of PBMCs. They could induce PBMCs to produce IL-2 but not IFN-r.They also could stimulate PBMCs to express IL-2R. Zhuling-duotang and BCG did not affect the percentage of CD, CD,CD cells among PBMCs. (2) HB vaccine did not significantly increase the cytotoxicity of PBMCs. (3)The cytotoxicity of PBMCs induced by ZhLling-daotang combined with HB vaccine was not significantly higher than that of PBMCs induced by Zhuling-duotang alone.
