自体血管内皮细胞种植人造血管的实验研究 被引量:1

Exlperimental study of implantation of autologous vas-cuiar endothelial cells on dacron grafts

摘要 用内径4mm,长5cm的涤纶人造血管对10条犬的17侧颈动脉进行了移植术。实验组A的人造血管种植了用0.25%胰蛋白酶自体颈外静脉获取的内皮细胞(n=7),实验组B种植了在液氮中冻存一周的自体颈外静脉的内皮细胞(n=3)。未种植内皮细胞的人造血管移植于实验组A的对侧颈动脉(。=7)作为对照组。术后不同时间取材,实验组A的总通畅率为85.7%,对照组为57%;实验组A的血栓量为40.6±39.6mg,对照组为85.9±26.3mg(P<0.01)。扫描电镜检查人造血管中段可见椭圆形细胞,而对照组仅见血小板、纤维素覆盖。透射电镜、VIII因子相关抗原以及Vimentin染色均证明人造血管中段细胞是内皮细胞。我们认为人造血管即刻种植酶解法所得的内皮细胞或种植在液氮中冻存一周的内皮细胞都能促进其内皮化,达到改造人造血管质量及性能的目的。 AbstractTen mongrel dogs underwent carotid artery by-pass 17 times with dacron grafts,4mm in inner diame-ter and 5cm in length.The grafts in experimentalgroup A were immediately seeded with autologousendothelial cells harvested by 0.25%trypsin solution(n=7).The grafts in experimental group B Wereseeded with autologous endothelial cells cryopreservedin liquid nitrogen for l week(n=3).The grafts incontrast group was not seeded with any endot helialcells implanted in the contralateral carotid arteries ingroup A (n=7).The number of endothelial cells har-vested in the two groups was 0.9±0.3 × 10.FactorVIII related antigen stain method confirmed the cellsharvested by 0.25%trypsin solution to be endothelialcells。The grafts were rernoved and studied at the endof 2,4,and 6 week after implantation. The total paten-cy rate of group A was 85.7%(6/7),and that of thecontrast group 57%(4/7).The weight of thrombus ingroup A was 40.6±36.9mg and the contrast group85.9±26.3mg(P<0.01 ).Scanning electron mi-croscopy revealed that the cells at the middle segmentof the grafts in the group A and B had the chracteris-tics of ellipse,and tight intercellular junction,whilethe middle segment grafts in contrast fIroup were covered by platelets,erythrocytes,leukocytes and fibrin。Transmssion electron microscopy, factor VIII relatedantigen slain method,and vimentin stain method de-monstrated that the cells in the group A and B showedcharacteristics of endothelial cells。 It was concludedthat endothelialization of the dacron grafts could be ac-celerated and the quality could be irnproved by immedi-ate seeding with endotheliai cells or by seeding withendothelial cells cryopreserved in liquid nitrogen for 1 week。
