摘要 研究Fe_3O_4超微粒子的毒性及其在小鼠体内的分布和排泄状况。结果:静脉注射Fe_3O_4超微粒子的小鼠半数致死量(LD_(50))为2022mg/kg体重。犬长期毒性试验观察到,血常规、肝、肾功能均在正常值范围内。心电图显示,在短时间内,心率有一过性增高,其间Q-T间期缩短,少数犬伴ST段轻度下移,但P-QRS-T波群未见明显异常改变。病理检验发现,肝、脾有不同程度肿大,其组织未见坏死。采用同位素示踪法证实,小鼠体内Fe_3O_4超微粒子的主要分布依次为肝、脾、肺、肾等脏器。其排泄途径以肾脏为主,56天经肾排出注射总量的56.8%,经肠道排出24.1%。半排期为17天。 Toxication,distribution,and excretion of Fe3O4 ultramicron afteri..infection were studied in mice and dogs.The LD50 of mice was 2022mg/kg.In chronic toxication experiments observed in dogs,the blood routine, and hepatic and renal functions laid within normal limit.ECG showed a transient tachycardia with a shorted Q-T interval.A slight drop of ST segment could be seen in a few dogs without significant change of P-QRS-T waves.In pathological examination.the liver and spleen had some enlargement but no necrosis was seen.with radioactive tracer,it was confirmed that Fe3O4 ultramicrons intensely distributed in liver,spleen,lung and kidney in mice in sequence.The main excretion approach was through kidney.excretiug 56.8% of the injected amount 56 days after injection,24.1% being through digestive tract.The half excretion time was 17 days.
