摘要
艾滋病(AIDS)的大流行,仍在全球迅速蔓延,极大地危害人类的生命及健康,临床迫切需要寻找新的抗人免疫缺陷病毒(HIV)药物,小动物模型是发展新药的重要环节,我们引进C型鼠白血病病毒(Ra-MuLV-RVB-3,RVB-3),建立了Balb/C小鼠感染模型,观察了RVB-3的感染特征。用AIDS首选药──叠氮胸苷(AZT)及黄芩提取品(SBT),在该模型内进行实验治疗,两者在体内实验中均显示明显的抗病毒效果。
The epidemic of the acquired immunodeficiency syndrome(AIDS),caused by the human immunodeficiency virus type I(HIV-I), requires rapid development of effective therapy and prophylaxis. A Practical small animal model system for HIV-I, in which candidate anti-HIV drugs could be evaluated rapidly for pharmacokinetics, toxicity and antiviral efficacy,would facilitate the selection of drugs for clinical trials.We import type C murine leukemia virus-Ra-MuLV-RVB-3 and set up a mouse model by Balb/C mice. The characteristics of infected mice have been observed. 3'-azido-3' deoxythymidine(AZT) and Scutellaria baicalensis tablets(SBT) have been evaluated in this model. AZT(30mg/kg, PO,bid x 10)significantly reduced spleen index(SI) by73. 3% and the WBC count by 47.4%. Three dose groups of SBT(5.0g/kg,2.5g/kg,1.25g/kg,PO,bid x 23)reduced SI by 53.0%,23.0% and 12.0% respectively and WBS count by 75.4%, 41.6% and 43.6% respectively.Easy handling,cheap cost and short term of experiment are major advantages of this model for anti-HIV drug evaluation.
