摘要
采用高效液相色谱——电化学检测法测定纹状体内单胺类递质多巴胺及其代谢产物3,4一二羟基苯乙酸,去甲肾上腺素含量,利用放射配体结合分析法检测多巴胺D_2受体密度及亲和力,以研究肾上腺髓质(AM)脑内移植后帕金弥补单胺类神经介质缺乏,促使超敏的D_2受体下调近至正常水平。结果提示AM脑内移植后的神经再支配可能是移植物长期发挥作用的机制,AM脑内移植是治疗PD的有效手段。
In order to investigate the intrastriatalneurochemical
changes of Parkinson’s disease(PD) following intracerebral implantation of a-drenal
medulla(AM)and demonstrate themechanism of grafts of AM curing parkinson-ism,the levels of
the neurotransmitter of dopamine and its metabolite,3,4-dihydrox- yphenylacetic acid and
noradrenaline,werequantitatively measured by HPLC- ECD. Meanwhile,the densities and the
affinities ofdopamine D_2 receptors were also detected byusing the method of radioligand
binding as-say. It was found the grafts of AM could al-mostly provide a substantial restoration of
thedopaminergic neurotransmitters in the previ-ously denervated striatum,make the
supersen-sitivity of the densities of dopamine D_2 recep-tors down-regulation closed to normal
leveland recover the motor asymmetry of PD rats.Our observations reveal that
intracerebraltransplantation of AM grafts is an efficientway for the tneatment of PD .The
evidences imply that the renervation between the hostand the grafts are the possible basic
mecha-nisms for the improvements.
