摘要
小鼠B—杂交瘤7TD1细胞的生长依赖于IL—6的存在。本研究对有或无IL-6条件下,对7TD1细胞的生物学行为加以分析。结果发现:1.细胞在缺乏IL—6时,生长停滞并出现细胞凋亡的典型特征;2.DNA核酸酶抑制剂ATA可有效防止因IL—6缺乏所致的凋亡;3.RNA合成抑制剂CHX能充分抑制IL—6对7TD1的生物效应,从而使细胞步入凋亡。本研究提示,IL—6通过抑制7TD1内部持续存在的细胞死亡程序来促进细胞存活和生长;而这种抑制作用需要新的蛋白质合成。
Murine B-hybridoma 7TD1 cells depend upon IL-6 for their growth and survival. In this study,the biological behaviors of 7TD1 cells under the conditions with or without IL-6 supplementation were investigated. The results showed 1)cell growth arrested and typical apoptotic patterns appeared when IL-6 had been deprived ;2)the endonucle-ase inhibitor ATA could prevent apoptosis induced by IL-6 deprivation and 3)a RNA synthesis inhibitor,CHX,could suppress the bioactivity of IL-6 on 7TD1 cells and push the cells to enter into apoptotic pathway. Our data thus,for the first time,suggest that IL-6 could promote cells survival and proliferation through the inhibition of a consistent suicide program inside 7TD1 cells. And such an inhibition might necessitate de novo protein synthesis.
