摘要
选用30只成年Wistar大鼠,探讨于坐骨神经损伤局部应用神经生长因于(NGF)及NGF抗体对脊神经后根节神经元的保护作用。结果表明,坐骨神经切断性损伤将引起29%的感觉神经元死亡。于损伤局部连续施用NGF可保护98%的感觉神经元在周围神经损伤后1月存活,而施加抗NGF抗体会进一步使胞体死亡率增至36%。因而对损伤性神经元死亡的保护,在临床也会有积极的意义。
AbstractIt is well establlshed that certain neurons undergodegenerative changes and die following peripheralnerve injuries.In view of the fact that neuronal cellbody exercises a major influence on nerve regenera-tion,the preqent study atternpts to provide a solutionto the protection of axotomy induced sensory neuronloss. 30 adult Wistar rats were used in this experi-ment.The neuronal cell counts were determined up toone month after sciatic nerve transected.Injured ma-ture sensory neurons treated locally with nerve growthfactor at the site of axotomy were almost completelyprotected from cell death(survival rate 98%,as com-pared with the death rate of 29%caused by sciaticnerve transectiOn), while local admimistration of anti NGF led to a further increment of neuronal celldeath (death rate 36%).Clinical application of NGF inprotecting axotomy induced sensory neuron fromdeath was discussed.
