摘要
应用一期法测定了16~22周胎儿肝、脾、肾组织中凝血因子VIII:C(FVIII:C)活性,发现肾组织中FVIII:C活性为1.723IU/ml,高于肝和脾(P<0.01)。三种组织细胞培养液中肾细胞培养液的FVIII:C活性始终较高。LSAB法免疫组化检测发现在肾远曲小管和集合管上皮细胞以及肝的胆管上皮细胞和肝、脾血管内皮细胞FVIII:CAg呈阳性。逆转录-多聚酶链反应证实三种组织中均有FVIIImRNA表达。表明在16~22周胎龄期肝、脾、肾均可合成FVIII:C,其中以肾最为明显。
actor VIII coagulation activity (FVIII :C ) in theliver, spleen and kidney of 16-to 22-week-old fetuseswas assayed with onestage method. FVIII :C in kid-ney was 1. 679 IU/ml , which was higher than that inspleen and liver (P<0. 01). FVIII :C was also higherin renal cell culture medium than in the other two or-gans. Epithelial cells of the following fetal tissues wereimmunohistologically (LSAB) positive for FVIII: CAg: distal convoluted renal tubules, renal collectingtubes and biliary tubes. FVIII:C Ag was also positivein endothelial cells of blood vessels in liver and spleen.By using reverse transcription-polymerase chain reac-tion (RT-PCR) technique, it was found that FVIIImRNA was expressed in liver, spleen and kidney all.The above results suggest that in 16 to 22-week-old fe-tuses the liver, the spleen and especially the kidneysynthesize FVIII : C.
