脐血长期冷冻保存及其冷冻损伤可恢复性的研究 被引量:6

STUDY ON LONG TERM CRYOPRESERVATION ANDHEMATOPIETIC PROGENITOR RECOVERY IN CORDBLOOD

摘要 作者应用二步法造血细胞冷冻技术,对脐血造血细胞在液氮中的长期保存的效果作了研究。结果表明:脐血造血细胞在低温保存时有一部分细胞会受到冷冻损伤而失去增殖能力,但是这种损伤并不与保存时间成正比。同时,我们还对脐血造血细胞冷冻损伤可恢复性作了研究。实验表明:在细胞悬液复温后即加入重组人粒-巨噬系集落刺激因子(rhGM-CSF)预孵育3小时后,可明显提高冻存人脐血造血细胞集落产率,这说明部分造血细胞在一定条件下可恢复其增殖能力,对应用冻存脐血作骨髓造血重建有一定意义。 he serum-free culture systern of hematopoieticcell was used to study the recovery rate of BFU-E,CFU-GM and CFU-GEMM in human cord blood atregular intervals of 3, 6 and 10 months after cryop-reservation (by two-step freezing technique). The re-sults showed that the cryoinjury mainly occurred in theprocess of freezing , but not in the course of cryopreser-vation. It was also found that the recovery rate ofBFU-E, CFUGM and CFU-GEMM was increasedwhen mononuclear cell suspension was incubated withrhGM-CSF without centrifugation. This suggestedthat some cryoinjuried progenitors may regain theircapacities of proliferation under certain conditions andthat this will be of great importance in utilizing cryop-reserved cord blood as grafts to reconstitute bone mar-row hematopoiesis .
