PROLIFERATION AND DIFFERENTIATION OF SINGLE CELLS

摘要 为深入探讨细胞亚群的不均一性及单个细胞的增殖分化特性,利用FACSVantage自动细胞分选系统(ACDU)分选出单个亚群细胞:和,在96孔扳无血清培养体系中,经SCF、IL-3、IL-6、GM-CSF、G-CSF和Epo.刺激,14~18天后扩增形成原代集落。单个细胞形成原代集落的比例为38.O%±9.0%,集落组成细胞趋于分化成熟,再接种培养,仅8.5%±2.5%可形成次级集落,且绝大部分为粉巨噬系细胞集落(CFU-GM)。54.0%±12.O%的单个细胞可形成原代集落,组成细胞形态均一,大多为原始幼稚细胞,75.0%±11.0%的原代集落再接种可形成次级集落,且产率高达280.O±21.5/原代集落,并可向红、粒、巨噬、巨核等系分化。表明细胞群的不均一性,其亚群显示造血祖细胞性能,而细胞的性能则接近于造血干细胞。 ingle and CD_(38) cell sorting wasperformed on a FACS Vantage by using the AutornatedCell Deposition Unit (ACDU). Both the subpopulations were sorted as single cells into 96-well platescontaining serumwfree culture medium supplementedwith SCF, IL-3 . IL-6,GM-CSF,G-CSF and Epo. Anaverage of 38%± 9 % of cells and 54%±12% of cells gave rise to an expansion ofcells after 14 ~ 18 day cell culture. The primarycolonies from single cells contained moredifferentiated myeloid cells, but those from single cells consisted of blastlike cells. The re-plating capacities of primary colonies of and cells were 8. 5% ±2. 5% and 75 %±11%, respectively. Most of the second generationcolonies from cells were CFUGM , but cells could give rise to CFUGM, CFU-E, BFU-E and CFU-GEMM in their second generationcolonies. These results indicate that cell popula-tion is still heterogeneous and its subsets. and CD_(38) , represent the behaviors ofhematopoietic progenitor cells and stem cells, respec-tively.
