摘要
为消除肌酐测定中血清胆红素干扰,采用由卫生部临床检验中心提供的高纯度胆红素氧化酶(BOX)和苦味酸试剂,设计了在COBASFARA-Ⅱ型全自动生化分析上检测肌酐的新方法,仅需样品8μl,BOX8μl,苦味酸试剂80μl,在190秒钟就可完成反应,所得结果表明,用该法对75份总胆红素在0~616μmol/L的标本进行检测,可以有效的消除胆红素对肌酐测定时的干扰。而且结果准确可靠,回收试验,黄疸血清标本用原法回收率仅为49%~71%,而用本法回收率可达94%~105%。本法的重复性也较好,批内变异系数(CV)为2.8%~12.7%,批间CV为14.0%。本法操作简单,标本也不需要作人工预处理,是肌酐测定消除胆红素干扰比较理想的方法。
he main interference while using Kinetic Jaffe’s method for the determination of creatinine was bilirubin;it made the results of creatinine inaccurate,lower and even negative in the samples of high bilirubin level.We selected high purity reagent of bilirubin oxidase(BOX) and ascorbatic acid,designed a new procedure on COBAS FARA-Ⅱ analvser to establish new method for determination of creatinine in jaundice patients. It only needs 8μl sample,BOX 8μl,Ascordatic acid 80μl and takes 190 seconds.The resul t was accurate,and indicated that the new method is free from interference or bilirubin. This delection method was used in 75 samples,which T Bil ranged from 0~616μmol/L. The result is accu-rate and reliable. The repetition is good.The CV of intra-group was 2.8%~12.7%,the CV of inter-group was 14.0%,The rate of recovery was 94%~105%. While the old method was 49%~71%. It is convenient to manipulate and the samples need no preparation. It is an idealistic method to determin creatinine deminishing bilirubin interference.
