摘要
为判断乙型肝炎临床阶段和预测患者预后,以PreS_1(P_(21~47))合成肪抗原包被酶联板、辣根过氧化物酶标记的羊抗人(兔或鼠)IgG为第二抗体建立酶联免疫吸附法(ELISA)检测PreS_1抗体。结果,PreS_1合成肽抗原可以检测到PreS_1合成肽免疫血清及乙型肝炎患者血清中PreS_1抗体;中和试验结果提示这种抗体可被PreS_1合成肽中和,中和抑制率大于70%,而不被血蓝蛋白、戊型肝炎病毒合成肽及丙型肝炎病毒合成肽中和。用Dane颗粒和基因表达PreS_1蛋白为抗原也可以检测到PreS_1合成肽免疫血清中的PreS_1抗体。结果表明PreS_1抗体力一保护件抗体,反映肝细胞损伤和病毒清除,其阳转时间越短,急性乙型肝炎病程就越短(r=0.67,P<0.001),示预后良好。
o meet the need of diagnosis of hepatitis B clinical stage and to predict its prognosis, an ELISA kit was developed to detect the anti-PreS1 antibody, using PreS1(21~47aa)synthetic peptide as coatedantigen,and the peroxidase labelled anti-human(rabbit or mouse)IgG as the second antibody. The re-sults show that the anti-PreS1 antibody was detected in the PreS1 synthetic peptide immune serum andhepatitis B patient’s serum,which were neutralized by PreS1 synthetic peptide and their neutralizationinhibition rate ranged from 70%~90%. They could not be neutralized by the synthetic peptide of HEVand HCV and keyhole limpet hemocyanin(KLH).The similar results also occurred in using Dane parti-cle and gene expression PreS1 protein as antigens to detect the anti-PreS 1 antibody in PreS_1 syntheticpeptide immune serum. Anti-PreS1 reflects both the hepatocellular injury and virus clearance.The earli-er anti-PreS_1 antibody shift positive,the shorter acute heptatitis B patient's course was.
