摘要
双糖铁培养基在鉴定革兰阴性杆菌中很重要。而某些国产双糖铁培养基上,细菌的硫化氢试验和氧化发酵试验不正确。经选择部分菌种对4个厂家生产的双糖铁培养基进行评价,认为培养基的pH是个关键。在pH7.5时双糖铁培养基才能正确反映上述两种试验的结果。建议选择硫化氢阳性的大肠埃希菌、弗劳地枸橼酸杆菌、腐败假单胞菌等针对硫化氢试验,不动杆菌针对氧化。发酵试验对双糖铁培养基进行质量控制。
he pH of kligler iron agar (KIA )effects H_2S reaction and oxidation-fermentation phenomenon of gram-negtive rods. We studied several local KIA products and found that there was difference in pH.By means of H_2S weak-producing strains we compared the H_2S reactions at pH from 6.4~8.0.It was found that the optimal pH of KIA was 7.5,where the medium gave a good O-F reaction and avoided H_2S false negtive results.The quality control strain for H_2S should be H_2S-positive E coli, C freundii and P putrefaciens which gave weak reactions.Acinetobater was also included to control O-F test.
