摘要
研究了三种维甲酸同分异构体,即全反式维甲酸(ATRA)、9顺式维甲酸(9C-RA)和13顺式维甲酸(13C-RA)在体外对人急性早幼粒细胞白血病(APL)细胞株NB4细胞的增殖和分化作用。形态学、细胞生长曲线、细胞周期动力学、细胞膜表面分化抗原、硝基蓝四唑试验(NBT)等多参数研究的结果表明,维甲酸诱导细胞分化早期伴有细胞的增殖;ATRA诱导NB4细胞分化的效应优于13C-RA,而9C-RA又优于ATRA。这对阐明维甲酸诱导早幼粒白血病细胞分化的作用机理,对发掘具有应用前景的新型维甲类分化诱导剂具有一定的价值。
e examined the effect of three RA isomers. in-cluding ATRA , 13C-RA and 9C-RA , on the prolifer-ation and differentiation properties of NB4 cells, a cellluie established from APL patient and carrying thetypical translocation t(15 ; 17 ). Standard parameterssuch as cell niorlihology, cell growth curve. dynamicsof cell cycle, expression of clusters of differentiationand reduction of nitro blue tetrazolium (NBT) wereused to evaluate the effects of the three ison iers. Dur-ing the first 48 hours of RA treatment, the APL cellmaturation was coupled with the cell prohferation.Moreover, significant differences of differentiation-in-duced effect aniong RA isomers were observed. ATRAshowed beuer results than 13C-RA while 9C-RA func-tioned even better than ATRA.
