摘要
作者采用荧光染色法检测,证实人胃癌SGC-7901细胞具有雌激素受体。在细胞培养过程中予以不同浓度的雌二醇处理,结果显示:于0.001~1μmol/L浓度时可刺激SGC-7901细胞的生长,其中以0.1μmol/L刺激作用最强,细胞群体倍增时间短于对照组(P<0.05),克隆形成率明显高于对照组(P<0.01),c-myc癌基因的表达亦较对照组强。流式细胞术检查发现,经雌二醇处理后,细胞增殖指数略高于对照组,而雌激素受体抑制剂三苯氧胺,可部分抑制雌二醇所产生的上述这些效应。实验结果提示,生理浓度的雌二醇对SGC-7901细胞具有一定的生长刺激作用,而三苯氧胺可部分抑制这种刺激作用。其机制可能是通过雌激素受体介导的。
Abstract In this study,the effect
was studied of estradiol(E_2)and tamoxifen on the growth of a hu- man
gastric adenocarcinoma SGC-7901 cell line in vitro. Estrogen receptor
as revealed by immunohistochemical staining was positive. when added
at various concentrations.E_2 could significantly stimulate cell
growth.The effect was most marked at concentration of 0.1 μmol / L.
The cell doubling time was reduced,the plating efficiency was
increased and c-myc oncogene expression was enhanced. Flow cytometric
analysis indicated that cell proliferation index was greater in the
E_2-treated than in the control.All these effects caused by E_2 could
be partially inhibited by the estrogen receptor inhibitor tamoxifen.
