摘要
作者采用MTT比色法,测定了32例体外原代乳癌细胞对6种常用化疗药物的敏感性。结果表明,不同乳癌对同一化疗药物的敏感性不同,不同乳癌有其各自的药敏谱。由于本组瘤群的IC_50_呈明显的偏态分布(P<0.0001),按IC_50的大小,可用百分位数法将其敏感性分为四类。初步临床分析表明,不同化疗药物的体外敏感性与治疗效果基本相符,提示本分析法可作为优化个体化疗的有效途径。
AbstractChemosensitivities of 32 human breast
cancers to 6 antitumor drugs were assayed in vitrousing MTT
colorimetric method.The results showed that the chemosensitivity of
primary breastcancer cultures varied remarkably from one patient,and
from one drug,to another with hun-dred-to thousand-fold
differences.The distribution of 50% inhibition concentrations
(IC_50)was significantly deviated from normal distribution(P<0.0001),
and the drug sensitivity couldbe divided into four different
catagories by means of percentile method. Preliminary resultsshowed
that the in vitro chemosensitivities of different antitumor drugs
were consistent with theclinical therapeutic responses It seems that
the herein reported assay might be useful on an indi-vidual basis to
optimize chemotherapy of human breast cancer.
